Do a combination of ferry and flight from Iriomote-jima to Yakushima-cho in 10 hours. Traveling from Iriomote-jima in October, expect Yakushima-cho to be slightly colder, temps between 30°C and 23°C. Finish up your sightseeing early on the 8th (Sun) so you can catch the flight to Nagasaki.

On the 10th (Tue), you'll have a packed day of sightseeing: snap pictures at Megane Bridge, explore the world behind art at Nagasaki Prefectural Art Museum, then steep yourself in history at Glover Garden, then walk around Nabekanmuriyama Park, and finally take your sightseeing to a higher altitude at Mt. Inasa.
