Virtually unchanged for the last six centuries, Venice is actually a group of islands made famous by a series of canals, bridges, monuments, piazzas, and narrow pedestrian lanes.
You've added San Giorgio Maggiore, Chiesa di San Zaccaria, Canal Grande, and other attractions to your itinerary. Do some cultural sightseeing at Basilica di San Marco and Santa Maria dei Miracoli. You've added Palazzo Vecchio, Cappelle Medicee, Mercato Centrale, and other attractions to your itinerary. On the 22nd (Wed), contemplate the long history of Colosseum, then don't miss a visit to Roman Forum, and then explore the ancient world of Palatine Hill. Get ready for a full day of sightseeing on the next day: steep yourself in history at St. Peter's Basilica, explore the galleries of Sistine Chapel, admire the masterpieces at Vatican Museums, then explore the world behind art at Galleria Borghese, then admire the local landmark of Trevi Fountain, and finally admire the landmark architecture of Pantheon.

You can take a train from Florence to Rome in 2 hours. Other options are to drive; or fly. Traveling from Florence in May, plan for a bit warmer nights in Rome, with lows around 58°F. You will have some time to spend on the 25th (Sat) before leaving for Sorrento.
